Figure 1: FACS: Human PBMCs were cultured with recombinant IL-1β, TGF-β, and IL-6 for 4 days. Cells were then reactivated with PMA/Ionomycin in the presence of monensin for 5h. Cells were stained with anti-IL-17A (human), mAb (17-142F) (FITC) (Prod. No. AG-20A-0071F) and PE anti-CD4. Cells in the lymphocyte gate were used for analysis. IL-17A exhibits multiple biological activities on a variety of cells including the induction of IL-6 and IL-8 production in fibroblsts, the enhancement of surface expression of ICAM-1 in fibroblast, activation of NF-κB and costimulation of T cell proliferation. Induces stromal cells to produce proinflammatory and hematopoietic cytokines.
