Figure 1: Immunofluorescence staining (green) of cryo-sections of human foreskin (fixed 15 min in 4% PFA) using anti-PROX1, pAb (dilution 1:200) (Prod. No. AG-25T-0105) and counter staining of nuclei with Dapi. A) Specific green PROX1 signal in the nuclei of lymphatics (L). Ep=epidermis.
Figure 2: Immunohistochemical detection of endogenous PROX1 (paraffin sections) in equine mandibular lymph nodeusing anti-PROX1, pAb (dilution 1:200) (Prod. No. AG-25T-0105), counterstained with toluidine blue. Sinusoids (S) as well as lymphatic vessels in the hilum of the lymph node (arrows) are lined by positively stained lymphatic endothelial cells. Blood vascular endothelium remains unstained (arrow heads). LC=Cluster of lymphocytes.

Description
Expression of PROX1 in blood endothelial cells induces expression of other lymphatic marker genes. Together with podoplanin, PROX1 can be used to reliably distinguish lympathic vessels from blood vessels. PROX1 is expressed in CNS, eye, pancreas, liver and heart, and it is one of the most specific and reliable markers for lymphatic endothelial cells.
