SLU-PP-332 is a potent and selective pan-estrogen-related receptor (ERR) agonist (EC50 values are 98 nM, 230 nM and 430 nM for ERRα, ERRβ and ERRγ respectively).

SLU-PP-332 increases energy expenditure and fatty acid oxidation, decreases fat mass, and improves insulin sensitivity. It enhances mitochondrial function and cellular respiration in skeletal muscle cell lines.

SLU-PP-332 decreases body weight and fat mass, blood levels of total cholesterol, HDL and triglycerides and adipocyte size in a mouse model of high-fat diet-induced obesity when administered at a dose of 50 mg/kg twice per day. It reduces blood glucose levels in a glucose tolerance test in the same animals at the same dose.

Estrogen-related receptors ERRα and ERRγ are essential regulators of cardiac metabolism. SLU-PP-332 significantly improved ejection fraction, ameliorated fibrosis and increased survival associated with pressure overload-induced heart failure (HF) without affecting cardiac hypertrophy.
