SMARCAL1 is a ATP-dependent DNA annealing helicase involved in maintaining genome stability. It plays a key role in replication fork repair and restart, helping resolve stalled replication forks to prevent DNA damage and ensure proper DNA replication. SMARCAL1 binds selectively to fork DNA relative to ssDNA or dsDNA and catalyzes the rewinding of the stably unwound DNA. It rewinds single-stranded DNA bubbles that are stably bound by replication protein A (RPA). It acts throughout the genome to reanneal stably unwound DNA, performing the opposite reaction of many enzymes, such as helicases and polymerases, that unwind DNA. Mutations in SMARCAL1 are associated with Schimke immuno-osseous dysplasia (SIOD), a rare multisystem disorder.
