Description
PicoGreen is a highly sensitive fluorescent dye specifically designed for the quantitation of double-stranded DNA (dsDNA) in solution. It exhibits exceptional selectivity, binding preferentially to dsDNA over single-stranded DNA, RNA, or free nucleotides, thereby minimizing background interference. Upon binding, PicoGreen produces a strong fluorescence signal with excitation/emission maxima around 480/520 nm, enabling detection of DNA at concentrations as low as picograms per milliliter. When PicoGreen binds to double-stranded nucleic acids, its fluorescence intensity increases approximately 1000-fold compared to unbound PicoGreen. This makes it an indispensable reagent in molecular biology for applications such as qPCR, DNA quantification in genomic and plasmid preparations, next-generation sequencing workflows, and high-throughput screening.
