DLK1 (mouse):Fc (human) (rec.)
Method: A mouse preadipocyte cell line, 3T3L-1, was stimulated with 5μg/ml of mouse DLK1:Fc as shown in indicated time points and each cell lysate was prepared and subjected to western blot by using an anti-Sox-9 and anti-GAPDH antibody.
Method: 3T3L-1 cells were maintained in DMEM, supplemented with 10% fetal bovine serum, penicilin-streptomycin and glutamine. For differentiation of 3T3L-1 cells, they were cultured in adipogenic medium which was growth medium supplemented with 1μM dexamethasone, 0.5mM IBMX, 10μg/m insulin (day 0). Medium was changed every 3 days. Staining with Oil Red O was typically performed on day 7. Cells were washed twice with PBS, fixed with 3.7% formalin and stained with 0.5% filtered Oil Red O in propylene glycol. Mouse DLK1:Fc (5μg/ml) dissolved in DPBS was added to the differentiation medium. Plates were then used to differentiate 3T3L-1 cells.

The transmembrane and secreted protein delta-like 1 homolog (DLK1; Protein delta homolog 1; Preadipocyte factor 1; Pref-1) belongs to the EGF-like family. It can be cleaved by a tumor necrosis factor-α converting enzyme (TACE) to release a soluble form. DLK1 plays important roles in regulating cell differentiation such as adipogenesis and osteogenesis. Membrane-bound DLK1 promotes hypertrophic myotube formation, whereas soluble DLK1 inhibits myotube formation. The soluble form of DLK1 inhibits preadipocyte differentiation, while membrane DLK1 represses preadipocyte proliferation. DLK1 is also associated with tumor invasion.
