Method: IL-23R (human) (rec.) (His) is coated on an ELISA plate at 1µg/ml overnight at room temperature. IL-23 (mouse):Fc (LALA-PG)-KIH (human) (rec.) (Prod. No. AG-40B-0248) or a control IL-2 (mouse) (monomeric):Fc (LALA-PG)-KIH (human) (rec.) (Prod. No. AG-40B-0264) is added (starting at 8000 ng/ml with a twofold serial dilution) during one hour and then detected using a human Fc (HRP) antibody. Note that IL-23R is only one subunit of the full IL-23 receptor that normally also includes IL-12Rβ1.

IL-23 is a heterodimeric cytokine containing two subunits: p19 (encoded by IL23A) and p40 (encoded by IL12B). IL-23 signals via a heterodimeric receptor, IL-23R, composed of a specific subunit, IL23R, and the IL12Rβ1 subunit, which is shared with the IL-12 receptor. IL-23R promotes mainly STAT3, but also STAT1, STAT4 and STAT5 phosphorylation. IL-23 supports the expansion and survival of established murine Th17 cells and enhances IFN-γ production by memory CD4+ T cells. IL-23R is expressed on T cells, B cells, myeloid cells and NK cells. IL-23 signaling pathway is important in the pathogenesis of several chronic inflammatory diseases, including axial spondyloarthritis, axial spondyloarthritis, Crohn's disease and psoriasis.
Cellular senescence is an age-associated mechanism that contributes to pathogenesis in diverse organ systems. In a recent study, it was observed that IL-23R, CCL5 and other proteins show age-dependent increases in circulation and are critical aging biomarkers, linked to senescence and are reduced by senotherapeutic agents. In human plasma, IL-23R abundance increases with age in both women and men. So, IL-23R seems to be a mediators of interorgan signaling mediated by senescent cells.
