Potent, selective, allosteric inhibitor of macrodomain 2 of poly-adenosine-diphosphate-ribose polymerase 14 (PARP-14 M2) (Kd=0.86µM, ITC (isothermal titration calorimetry) assay). PARP-14 is an anti-apoptotic pro-survival protein associated with inflammatory diseases and several types of cancer.

Engages PARP-14 MD2 in intact cells displaces adenosine diphosphate ribose (ADPR) from PARP-14 M2 and prevents recruitment of the PARP-14 M2 to DNA damage sites.
