Figure 1: 5x10^5 ficoll prepared human PBMCs were washed and pre-incubated 10 min with 20µl of 300µg/ml human IgG (to reduce non specific binding) after which they were incubated 45 min with 80µl of anti-TCR Cβ1/Biotin at 5µg/ml. Cells were washed twice and incubated with 50µl of 2nd reagent Strep/PE after which they were washed three times, fixed and analyzed by FACS. A 34% subpopulation of the cells stained positive with a mean shift of 2.09 log10 fluorescent units when compared to a Mouse IgG2a/Biotin negative control (ANC-281-030). Binding was blocked when cells were pre-incubated 10 min with 20µl of 0.5mg/ml anti-TCR Cβ1 antibody (ANC-101-020).

Recognizes human Cβ1 TCR gene product (TRBC1) and reacts with 50-75% of T cells in normal human blood. Does not bind to TCR Cβ2 (TRBC2).

Human T cell receptor (TCR) is a T cell surface expressed heterodimer of α/β or γ/δ chains. TCR’s recognize antigen peptides bound to MHC molecules forming the basis of antigen specific response by T cells. This specific TRBC1 antibody has been described to be useful in a simplified immunophenotypic assessment of T cell clonality.
